灯盏花对大鼠急性呼吸窘迫综合征细胞凋亡的影响

Effects of erigeron breviscapine on pneumocyte apoptosis during acute respiratory distress syndrome in rats

摘要 目的探讨灯盏花对大鼠急性呼吸窘迫综合征(ARDS)中细胞凋亡的作用及可能机制。方法建立大鼠在体ARDS模型,将32只大鼠按随机数字表法分为对照组(I组)、ARDS组(II组)、小剂量灯盏花组(III组,25mg/kg)和大剂量灯盏花组(IV组,50mg/kg),检测各组肺组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,应用免疫组化法检测bcl.2蛋白含量,DNA缺口末端标记法(TUNEL)检测肺细胞凋亡,光镜下HE染色观察病理形态学改变。结果与II组比较,III、IV组SOD活性及bcl-2含量明显增高,MDA含量、凋亡指数均明显降低,肺水肿程度显著减轻;III、IV组之间比较差异有统计学意义(P〈0.05)。结论灯盏花对ARDS具有防治作用,其机制可能与抗氧化、抑制氧自由基释放、上调bcl-2基因表达,从而减轻肺细胞凋亡有关。大剂量比小剂量保护作用更显著。 Objective To investigate the anti-apoptosis effects of erigeron breviscapine on acute respiratory distress syndrome (ARDS) in rats. Methods Thirty-two Sprague-Dawley rats were randomized into four groups with 8 animals in each group : (I) control group, (II) ARDS group, (III) erigeron 25 mg/ kg group, and (IV) erigeron 50 mg/kg group. An ARDS rat model was established by injection with hpopolysaccharide (LPS) 6 mg/kg; erigeron breviscapine was administrated 3 days before LPS injection. The lung tissue superoxide dismutase ( SOD), malondialdehyde ( MDA ), the content of bcl-2 ( immunohistochemical stain), and apoptotic cells (TUNEL) were detected. Pathological changes of lung tissues were observed with lightmicroscopy. Results The expression of bcl-2 and SOD activities of erigeron groups were significantly higher than those of ARDS group ( P 〈 0.01 ). The level of MDA in lung tissue of erigeron groups was lower than those of ARDS group ( P 〈 0. 01 ). Apoptotic index was significantly decreased after injection erigeron ( P 〈 0. 01 ). There was significant difference between group III and IV. Conclusions Erigeron breviscapine can reduce lung injury through enhancing anti-free-radical ability of the organism, upregulating the protein expression of bcl-2, and subsequently inhibiting pneumocyte apoptosis.
