摘要
目的:观察逍遥散及其功效拆方对CUMS模型大鼠血清白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)及皮质酮(Cort)水平的影响,初步揭示逍遥散中体现不同治法的功效拆方抗抑郁效应机制上的差异。方法:采用慢性不可预知温和应激+孤养6周的方法制备CUMS大鼠抑郁模型,逍遥散(30g/kg)、逍遥散中疏肝药(柴胡+薄荷,5.25g/kg)、逍遥散中养血药(当归+白芍,9g/kg)、逍遥散中疏肝药+逍遥散中健脾药(柴胡+薄荷+白术+茯苓+生姜+炙甘草,21g/kg)、逍遥散中养血药+逍遥散中健脾药(当归+白芍+白术+茯苓+生姜+炙甘草,24.75g/kg)、阿米替林(10mg/kg)连续4周灌胃给予(造模第3周开始给药),ELISA法测定大鼠血清IL-1β、IL-6、TNF-α、IFN-γ、Cort的含量。结果:与模型组比较,逍遥散、疏肝药、养血药、疏肝药+健脾药、养血药+健脾药均能显著降低大鼠血清IL-1β、IL-6、TNF-α、IFN-γ水平,且逍遥散、疏肝药、疏肝药+健脾药的作用优于养血药、养血药+健脾药;逍遥散、养血药、养血药+健脾药显著降低大鼠血清Cort水平,但疏肝药、疏肝药+健脾药的作用不明显。结论:逍遥散抗抑郁作用的发挥与抑制CUMS模型大鼠炎性细胞因子的释放、降低血清皮质酮水平有关,其中代表"疏肝"治法的柴胡-薄荷药组对细胞因子的调节作用突出,而代表"养血"治法的当归-白芍药组对皮质酮水平的影响作用显著,共奏全方抗抑郁效应。
Objective: To observe the effects of the decoction of Xiaoyaosan (XYS) and its efficacy drug-groups on the level of serum IL-1β, IL-6, TNF-α, IFN-γ and Cort in CUMS model rats, so as to reveal the difference between the mechanisms of anti-depression effect of various effica- cy drug-groups representing dissimilar therapeutic methods. Methods: Wistar rats were randomly divided into eight groups: XYS group (30g/kg), SG group (5.25g/kg), YX group (9g/kg), SG +JP group (21g/kg), YX +JP group (24.75g/kg), control group, model group, and amitriptyline group (10mg/kg). The depression rat model was established by using chronic unpredictable mild stressors and solitary feeding condition as described in the literature. From the third week of modeling, all rats were intragastrically administrated with different decoctions or distilled water for 4 weeks. The levels of serum IL-1β, IL-6, TNF-α, IFN-γand Cort were determined after the last administration by ELISA. Results: Compared with model group, XYS, SG, YX, SG + JP and YX + JP group all decreased the levels of serum IL-1β, IL-6, TNF-α, IFN-γ, and Cort, and the effects of XYS, SG and SG + JP group were more prominent than that of YX and YX + JP group. Moreover, the decoctions of XYS, YX and YX + JP could reduce Cort content in serum of model rats, while the significant effect was not performed by the decoctions of SG and SG -JP. Conclusion: The decoction of XYS shows antidepressant effect related to inhibiting the increase of serum cytokines and Cort. The SG drug-group plays a principal role in immune modulation and the YX drug-group acts as a regulator for endocrine system, both of them cooperate in antidepressant action of Xiaoyaosan.
