柚皮素对脂多糖诱导小鼠急性肺损伤的保护作用研究 被引量:17

摘要 目的:探讨柚皮素对内毒素(lipopolysaccharide,LPS)诱导小鼠的急性肺损伤(acute lung injury,ALI)保护作用。方法:于小鼠腹腔注射LPS(5mg/kg)复制诱导小鼠的急性肺损伤动物模型。将小鼠随机分为空白组、LPS组、地塞米松组(2mg/kg)、柚皮素高剂量组(40mg/kg)、柚皮素低剂量组(20mg/kg),观察各组肺组织病理学改变,测量肺湿/干重比,支气管肺泡灌洗液中性粒细胞百分比、SOD、肺组织中MPO及炎症因子IL-6、TNF-α含量。结果:柚皮素能有效减轻LPS所致肺组织病理学变化,降低肺泡灌洗液中炎症细胞的数目,显著降低肺湿/干重比重,提高SOD水平,并能降低MPO水平。结论:柚皮素可减轻LPS所致急性肺组织损伤,对LPS诱导的急性肺损伤有保护作用。 Objective: determine the protective effect of naringenin on acute lung injury induced by lipopolysaccharide (LPS). Methods: The BABL/C mice were randomly allocated into control, LPS, Dex(2mg/kg) + LPS group, naringenin(20mg/kg) + LPS group, and naringenin (40mg/kg) + LPS groups. LPS was intraperitoneally injected at a dose of 5mg/kg body weight in the LPS group, along with equal volumes of saline in the control group. Naringenin + LPS groups and Dex(2mg/kg) + LPS group were treated with naringenin ( 20mg/kg,40mg/kg, intraperitoneally) 1 h prior to the injection of LPS or saline, respectively. The mice were killed 6h after LPS challenge. The bronchoalveolar lavage fluid (BALF) samgles were analyzed for the concentration of numbers of neutrophilic,SOD,IL-6and TNF-α, and The lung samples were taken for histologic evaluation and for determination of wet-to-dry(W/D) lung weight, (MPO) level. Results: Pretreatment with naringenin prior to the administration of intratracheal LPS significantly induced a decrease in lung wet weight/dry weight ratio, in total leukocyte number and neutrophil percent in the BALF, and in MPO activity of lung in dose-dependent manners, and in IL-6, the tumor neurosis factor-α (TNF-α) in the BALF. At the same time, pretreatment with naringenin also significantly improved the superoxide dismntase (SOD) activity in the bronchoalveolar lavage fluid (BALF). These results showed that anti-inflammatory effects of naringenin against the LPS-induced ALl.
