甘草次酸对大鼠体内山柰酚组织分布的影响

摘要 目的:研究甘草次酸对大鼠体内山奈酚药物动力学的影响。方法:将78只SD大鼠,随机分成4组,雌雄各半,即高浓度实验组与低浓度实验组及对照组的高浓度组与低浓度组,每组中又包括三个小组即1h、2h、3h。高浓度实验组灌胃50mg/kg山奈酚+50mg/kg甘草次酸,低浓度实验组灌胃25mg/kg山奈酚+50mg/kg甘草次酸。对照组只灌胃等剂量的山奈酚。灌胃给药后分别于1h、2h和3h用摘眼球取血法取血,取血后立即处死大鼠,取出心、肝、肺、肾、脑组织,组织经处理后用HPLC法测定。结果:在肝脏、肾、肺、心中,灌胃山奈酚+甘草次酸后的组织样品的山奈酚含量与单独灌胃山奈酚的组织含量之间无显著差异。结论:甘草次酸对山奈酚在肝脏、肾、肺、心的分布没有影响。 Objective:To investigate the effect of grycyrrhetinic acid on the pharmacokinetics of kaempferol in rats. Methods :72 SD rats,weighing 140 - 200g, half male and half female, were distributed randomly into 4 groups ,including control group of high concentration and low concentration, experimental group of high concentration and low concentration. Then each group was divided into three sub-groups, namely group of lh, 2h and 3h. Experimental group of high concentration and low concentration were administered by gavage for a dose of 50 mg/kg kaempferol mixed with 50 mg/kg grycyrrhetinic acid and 25 mg/kg mixed with 50 mg/kg grycyrrhetinic acid, respectively. The control group were administered by gavage for a same dose of that of the experimental group except grycyrrhetinic acid. Plasma samples were collected by picking eyeballs lh, 2h and 3h after administration. Then the rats were sacrificed and heart,lung,kidney,liver and brain were collected. The concentration of kaempferol in tissues were monitored by HPLC. Results :The results show that at two concentration levels there is no significant difference (P 〉0.05) between two sets data of experimental group and control group in liver, kidney, lung and heart. Conclusion: grycyrrhetinic acid have no effect on the distribution of kaempferol in liver, kidney,lung and heart.
