解毒扶阳方对2型糖尿病大鼠代谢紊乱的影响

Effects of Jiedufuyang Fang( 解毒扶阳方) decoction on the metabolism disorder in type 2 diabetes mellitus model rats

摘要 目的:探讨解毒扶阳方对2型糖尿病(TZDM)模型大鼠代谢紊乱的调节作用。方法:取SPF级雄性大鼠,随机分为正常对照组、高脂高糖(模型)组、二甲双胍(降糖)组、立普妥(降脂)组、解毒扶阳方(试药)大、中、小剂量组。除正常对照组外,其他各组大鼠均以高脂高糖饲料喂养。除正常对照组外,各组大鼠禁食12h,按60mg/kg剂量尾静脉一次性快速注射链佐茵素制备动物模型,并于模型制作当日各给药组大鼠分别ig给予药物治疗,每日一次,连续12周。其中降脂组给予立普妥1.25mg/kg,降糖组给予二甲双胍670mg/kg,试药组给予解毒扶阳方浓缩液18g/kg、12g/kg、6s/kg,正常对照组、模型组给予同样量的生理盐水。于给药最后一天眼球取血测定空腹血糖;采用酶联免疫吸附分析方法测定血清瘦素、胰岛素、抵抗素、脂联素水平,计算胰岛素抵抗指数。结果:解毒扶阳方能调节血糖水平,与模型组比较,中、小剂量组有显著差异,大剂量组有非常显著性的差异。其瘦素含量与模型组比较,均显示有非常显著性的差异。能调节胰岛素水平,与模型组比较,均具有非常显著性的差异。对抵抗素、脂联素水平也具有明显的调节作用。结果表明,本方治疗T2DM的有效剂量为6g/kg,其有效范围为6~18g/kg。结论:解毒扶阳方对T2DM所致糖、脂代谢紊乱有调节作用。 Objective: To explore the regulatory effects of the Jiedufuyang Fang (解毒扶阳方)decoction on glycometabolism and lipometabolism disorder in type 2 diabetes meUitus(T2DM). Methods: 70 male specified-pathogens free(SPF) Wistar rats were divided into seven groups in randomized manner: the control group with distilled water diet; the model group with high fat ( lard 49% ) and high sugar ( edible sugar 10% ); the lipid-lowering group treated with liptor; the glycemic-lowering group treated with mefformin; the large dose, middle dose and small dosegroups treated with the decoction. The rats of every group were taken in high fat and high sugar and taken In addition to the normal control group, a group rats fasting 12h, intravenous single rapid injection preparation of animal model of streptozotocin 60mg/kg by except control group. In addition to the normal control group, other groups of rats were feeding with high fat and high sugar. In addition to the normal control group, other groups and model at all for set of rats given by ig drug treatment, once a day, for 12 weeks. Among them, the rats in lipid- lowering group were taken 1.25 mg/kg liptor; the rats in glycemie-lowering group were taken 670 mg/kg mefformin; the rats in the decoction large dose group, middle dose group and small dose group were taken 18 g/kg, 12 g/kg and 6 g/kg the decoction of concentrated solution separately. The rats in control group and model group were taken normal salt administration by garage for equal content. The blood glucose were determinated; the level of serum leptin, insulin, resistin and adiponeetin were detected by enzyme-linked immunosorbent assay (ELISA). And insulin resistance index (HOMA-IR) was counted. Results: Jie Du Fu Yang Fang decoction could regulate blood sugar lev- els, compared with model group, midium and small dose group had significant differences, high-dose group was very significant difference. Compared with model group, the leptin levels had shown there~ a very significant difference. Could adjust insulila levels, compared with mod- el group, had a very significant difference. Of resistin and adiponectin level also had the obvious adjustment. It was shown that the effective dose of the prescription is at 6g,/kg, and the effective range at 6 - 18g/kg. Conclusion: It was suggested that the decoction be of effects of regulation on metabolism disorder in saeeharide and fat as metabolic syndrome.
