摘要
目的:观察全身垂直振动对去卵巢骨质疏松大鼠全骨髓细胞和骨髓基质干细胞PPARγ和PGSK-3β蛋白表达的影响。方法:36只3月龄雌性SD大鼠按体重分层后随机分为假手术、去卵巢静止和去卵巢振动3组。去卵巢手术10周后,去卵巢振动组大鼠开始每天进行2次时间15 min、频率90 Hz的全身垂直振动治疗。振动治疗7周时,腹主动脉取血处死大鼠,迅速在无菌条件下截取双后肢,冲洗骨髓细胞,一部分细胞直接提取蛋白,另一部分细胞采用全骨髓方法培养后再提取蛋白。采用Western blotting方法检测提取蛋白中的PPARγ和P-GSK-3β蛋白表达水平。结果:(1)与假手术组比较,去卵巢静止组全骨髓细胞PPARγ蛋白表达无显著差异,而P-GSK-3β蛋白表达显著降低;与去卵巢静止组比较,去卵巢振动组全骨髓细胞PPARγ蛋白表达无显著差异,而P-GSK-3β表达显著增加。(2)与假手术组比较,去卵巢静止组骨髓基质干细胞PPARγ蛋白表达无显著差异,而P-GSK-3β表达显著降低;与去卵巢静止组比较,去卵巢振动组骨髓基质干细胞PPARγ和P-GSK-3β蛋白表达均无显著差异。结论:全身垂直振动能促进去卵巢骨质疏松大鼠全骨髓细胞P-GSK-3β蛋白表达,但其效应在体外培养后消失。
Objective This paper investigated the effects of whole-body vertical vibration (WBW) on PPARγ and P-GSK-3β protein expression in both whole bone marrow cells (BMCs)and bone marrow stro- mal cells (BMSCs) of ovariectomized rats with osteoporosis. Methods Thirty-six female SD rats were ran- domly divided into following three groups: sham-operation (Sham), ovariectomized (OVX), and ovariecto- mized plus vibration (OVX-V). After 10 weeks of operation, the rats in OVX-V group were treated with WBW at 90 Hz twice a day for seven weeks. Rats were sacrificed at the 7th week of WBW treatment, BMSs were flushed out from the hindlimb, and the proteins were extracted immediately from a part of the BMSs and from the remaining BMSs after whole bone marrow cell culture. The expression of PPARy (peroxi- some proliferators activated receptor)and P-GSK-3β (glycogen synthakinase)proteins was investigated by Western blotting. Results As compared with the Sham group, the expression of P-GSK-3β protein significant- ly decreased, but no significant change in the expression of PPARγ protein in whole bone marrow cells in OVX group. As compared with the OVX group, the level of P-GSK-3β protein significantly increased with- out significant change in PPARγ protein in OVX-V group. As compared with the Sham group, the level of PPARγ protein expression of BMSCs in OVX group changed insignificantly, while the P-GSK-3β protein de- creased significantly. As compared with the OVX group, the level of PPARγ and P-GSK-3 protein in OVX-V group changed insignificantly. Conclusion Whole-body vertical vibration could stimulate the protein expres- sion of P-GSK-3β in bone marrow cells, but this stimulating effect disappeared in cultured cells.
