摘要
目的探讨氢醌(HQ)对TK6细胞白血病相关miRNA表达的影响。方法以磷酸盐缓冲液(PBS)溶解HQ,以PBS组为对照组,分别以2.5、5.0、10.0和20μmol/L HQ染毒TK6细胞48 h。应用反转录实时荧光定量-聚合酶链反应检测miR-126、miR-155、miR-196b和miR-20b的表达改变。结果与对照组相比,HQ对TK6细胞的上述4种白血病相关miRNA的表达改变均有影响,miR-126的表达上调,miR-155、miR-196b和miR-20b的表达均为下调,且均以20μmol/L HQ组最为明显,miR-126、miR-155、miR-196b和miR-20b分别上调83%,下调40%,49%和42%(P<0.05)。结论 HQ能促进miR-126的表达,抑制miR-155、miR-196b和miR-20b的表达。
Objective To explore the expression changes of leukemia-related miRNAs induced by hydroquinone (HQ) in TK6 cells. Methods Hydroquinone was dissolved with PBS buffer, the TK6 cells were incubated with 2.5, 5.0, 10.0 and 20.0 μmol/L HQ for 48 h, respectively, and the cells treated with PBS were taken as the control. The expression of miR-126, miR- 155, miR-196b and miR-20b was detected by reverse transcription real-time RT-PCR assay. Results All the expressions of the above four kinds of miRNAs in HQ-treated cells were changed compared with the control group, the up-regulated expression of miR-126 and down-regulated expressions of miR-155, miR-196b, and miR-20b were observed, the 20 p.mol/L HQ treatment affected the expressions significantly, increased by 83%, and decreased by 40%, 48% and 42% respectively (all P〈0.05). Conclusion HQ may enhance the expression of miR-126, and inhibit the expression of miR-155, miR-196b and miR-20b.
