榭皮素对GSNO诱导损伤的内皮细胞的保护作用及其机制研究 被引量:1

摘要 目的:考察榭皮素(Quercetin)对S-亚硝基化谷胱甘肽(GSNO)诱导损伤的内皮细胞的保护作用,探讨其保护机制。方法:内皮细胞(EA.hy926)预先给予不同浓度榭皮素,之后给予GSNO损伤,建立模型。采用:噻唑兰染色(MTT法)检测细胞存活率测定,之后通过对超氧化物歧化酶(SOD),丙二醛(MDA),谷胱甘肽过氧化物酶(GSH-PX),活性氧(ROS),Caspase-3等指标测定进行分析。结果:榭皮素可明显提高内皮细胞内SOD,GSH-PX的活性,抑制MDA的产生,降低ROS的活性,减少细胞凋亡率,具有统计学差异(P<0.05)。结论:榭皮素对GSNO诱导损伤的细胞具有保护作用,其机制可能与榭皮素可提高细胞内抗氧化酶活性有关。 Objective :To investigate the protective effect of quercetin on endothelial cell injury induced by S -nitrosoglutathione, and its protective mechanism. Methods:Different concentrations of quercetin were given to endothelial cell ( EA. hy926) in advance,and then GSNO was treated to form the injury model. MTr method was used to value the cell viability. The expression of SOD, MDA, GSH - PX and ROS were analyzed. Results : The application of quercetin significantly protected the endothelial ceils from GSNO injury. The activation of SOD and GSH - PX was increased, and the expressions of MDA were down regulated by quercetin pretreatment. The generation of ROS was also significantly decreased. Finally, results in the percentage of apoptosis was down ( P 〈 0. 05 ) . Conclusion : Quercetin has protective effects on GSNO -induced apoptosis on endothelial cell, and the protective mechanism may enhance the antioxidant by quercetin.
