摘要
目的:建立同时测定保和颗粒中芦丁、橙皮苷和连翘苷含量的方法。方法:应用高效液相色谱法测定,色谱柱为AgilentZorbax SB-C18色谱柱(4.6mm×250mm,5μm);流动相为乙腈-0.05%磷酸溶液,梯度洗脱:流速为1.0mL·min^-1,柱温30℃;检测波长277nm。结果:芦丁、橙皮苷和连翘苷的线性范围分别为1.741-130.6μg·mL^-1(r=0.9992),1.687~126.5μg·mL^-1(r=0.9998),0.6304~47.28μg·mL^-1(r=1.0000);平均加样回收率分别为100.92%(RSD=1.05%),100.87%(RSD=1.45%),99.86%(RSD=1.73%)。结论:该方法同时测定保和颗粒中的芦丁、橙皮苷、和连翘苷的含量,简便易行、准确、重复性好,可用于保和颗粒的质量控制。
Objective :To establish a high performance liquid chromatographic method for simultaneous determinationthe contents of Rutin, Hesperidin and Phillyrin in Baohe Granules. Methods : HPLC was applied with the chromatographic conditions as follows:the chromatographic column was Agilent Zorbax SB -C 18 (4.6 mm × 250 mm,5 μm) at 30 ℃ ; acetonitrile -0.05 % H3 PO4 as the mobile phrase, gradient elution;flow rate was 1.0 mL . min^- 1, the detection wavelength was 277nm. Results : Linearity relationships were 1. 741 - 130.6 μg . mL^- 1 ( r = 0. 9992) , 1. 687 - 126.5 μg . mL ^- 1 ( r =0. 9998) ,0. 6304 - 47.28 μg . mL^-1 ( r = 1. 0000), respectively; the average recoveries were 100.92% ( RSD = 1.05% ), 100.87% ( RSD = 1.45% ) ,99.86% ( RSD = 1.73% ). Conclusion:The results showed that the method was simple, accurate and repeatable and it was suitable for simultaneous determination of the contents of Rutin, Hesperidin and Phillyrin in Baohe Granules.
