Box-Behnken设计优化超临界CO_2萃取丹葶肺心颗粒中挥发油的工艺研究

摘要 目的:优选超临界CO2流体萃取法萃取丹葶肺心颗粒中挥发油的工艺条件。方法:以萃取压力、萃取温度、萃取时间3个因素为主要影响因素,以鱼腥草素得率、鱼腥草挥发油得率、枳壳挥发油得率、总评归一值(OD)为评价指标进行Box-Behnken设计试验,考察最佳萃取工艺。结果:优化的最佳工艺条件为:萃取压力27.5 MPa,萃取温度58℃,萃取时间105 min,此时鱼腥草素、鱼腥草挥发油、枳壳挥发油得率分别为0.061%、1.973%,0.752%;对最佳工艺进行验证,结果实测平均值与模型预测值非常接近,表明该模型较可靠。结论:采用Box-Behnken设计方法科学、合理、可行;超临界萃取效率高,稳定性好,在中药复方挥发油的提取方面有相当大的发展潜力。 Objective :To optimize the extraction conditions of supercritical fluid extraction ( CO2 ) of volatile oil from Danting Feixin Granules by Box - Behnken design. Methods : In order to obtain the best extraction conditions,in the Box - Behnken design experiments, we studied the three principal factors of extraction pressure, extraction temperature and extraction time by the evaluation of yields of Decanoyl acetaldehyde, Houttuynia cordata Thumb oil and Citrus aurantium oil. Results:The optimized conditions were as follows:extraction pressure 27.5 MPa,extraction temperature 58 ℃ and extraction time 105 minutes. In such conditions, the yields of Decanoyl acetaldehyde ,Houttuynia cordata Thumb oil and Citrus aurantium oil were 0. 061% , 1. 973% and 0. 752% respectively. The results of the validation tests showed that the measured average value was consistent with the predicted value, and indicated that the model was reliable. Conclusion:The Box - Behnken design was scientific, reasonable and feasible. Supercritical fluid extraction had high efficiency and good stability and was considered to be the development potential technology using extraction of volatile oil from traditional Chinese medicine.
