摘要
目的:建立同时测定注射用丹心宁中丹参素、原儿茶醛、丹酚酸B、红花黄色素含量的反相高效液相色谱法。方法:以Agilent Zorbax Extend C18柱(250mm×4.6mm,5μm)为固定相,乙腈-0.03%磷酸为流动相.在0~65min,乙腈量从2%上升到33%,检测波长为280nm和403nm,流速0.8mL·min^-1。结果:4种成份分离良好,丹参素、原儿茶醛、丹酚酸B、红花黄色素进样量分别在范围0.313—3.756μg,0.053-0.6362μg.0.567—6.809μg和0.0443—0.5321μg线性关系良好,加样回收率分别为丹参素102.1%,RSD=1.1%;原儿茶醛97.3%,RSD=1.2%;丹酚酸B99.3%,RSD=1.2%;羟基红花黄色素A100.6%,RSD=1.5%。结论:本法简单、快捷、适合于测定注射用丹心宁中丹参素、原儿茶醛、丹酚酸B、羟基红花黄色素A的含量。
Objective :To develop a RP -HPLC method for the first time to simultaneously determine four major compounds in Zhusheyong Danxinning, namely danshensu, protocatechuic aldehyde, salvianolic acid B and hydroxysafflor yellow A with UV detection at 280 nm and 403 nm. Methods : The column was Agilent Zorbax Extend C 18 (250 mm × 4.6 mm,5 μm). The separation was carried out with a gradient program. The mobile phase was acetonitrile- 0.03% phosphoric acid at a flow rate of 0.8 mL . min^-1. Results :The standard curve was rectilinear in the range of 0. 313 -3. 756 μg ( r = 1. 00130) for danshensu ,0. 053 - 0. 6362 μg ( r = 1. (3000) for protocatechuic aldehyde,0. 567 - 6. 809 μg ( r = 1. 0000) for salvianolic acid B, and 0. 0443 - 0. 5321 μg ( r = 1. 0000) for hydroxysafflor yellow A. The recoveries of the markers listed above were 102.1% ,97.3 % ,99.3% , and 100.6% , respectively. Conclusion:The method is simple, rapid and suitable for the determination of four compounds in Zhusheyong Danxinning.
