摘要
目的 探讨 calponin基因对血管再狭窄的防治作用 ;评价用 infiltrator系统进行基因转染的实用价值 .方法 以p CAGGS质粒为基因载体 ,以脂质体为介导 ,用 infiltrator浸壁球囊导管向兔髂动脉壁内导入人重组 calponin基因 .结果 实验组 (n=6 )基因转染全部成功 . 4wk后转基因组血管内膜增生较轻 ,而对照组 (n=6 )血管内膜明显增生 ,管腔显著变窄 .未观察到与目的基因及转染系统相关的不良反应 .结论 infiltrator导管是活体局部转基因的合适工具 ;用该导管转染 calponin基因可能成为防治血管再狭窄的有效方法 .
AIM To investigate the preventive effects of calponin on restenosis of blood vessel and to assess the value of infiltrator system in gene transfection. METHODS The human recombinant calponin gene was transferred into the iliac artery walls of rabbits with infiltrator cathetor. pCAGGS and liposome were used as vector and mediator.RESULTS Intimal proliferation was significant in control group( n =6) but slight in transfection group( n =6) 4 weeks after transfection.No adverse effects associated with calponin gene or infiltrator system were observed.CONCLUSION The infiltrator catheter is a suitable device for local gene transfer in vivo ;calponin transfection with infiltrator system appears to be an effective method in prevention and treatment of blood vessel restenosis.
