摘要
目的 应用阿霉素 (ADR)建立心肌细胞中毒模型 ,观察牛磺酸 (Tau)对中毒心肌细胞的保护作用并探讨其机制。方法 将培养 72h后的新生大鼠心肌细胞分为对照组、ADR组和ADR +Tau组 ,继续培养 2 4h后用MTT法测定心肌细胞存活力 ,并测定细胞内丙二醛 (MDA)含量、超氧化物歧化酶 (SOD)活性及细胞内游离钙离子浓度 ([Ca2 +]i)。结果 Tau能够增强ADR中毒心肌细胞存活力 ,降低心肌细胞MDA含量及 [Ca2 +]i,同时升高细胞内SOD活性。结论 Tau能够对抗氧自由基引起的脂质过氧化和逆转细胞内钙超载 。
Objective To explore the protective effect of taurine (Tau) on toxic cardiomyocytes induced by adriamycin (ADR) and its mechanism. Methods The cultured neonatal rat cardiomyocytes were divided into control,ADR and ADR+Tau groups.After 24 hour further culture,the cardiomyocyte viability was analysed by MTT assay,the cell malondialdehyde (MDA) content,the activity of superoxide dismutase (SOD)in myocytes and the intracellular calcium concentration were determined respectively. Results Tau increased the viability of toxic cardiomyocytes and the activity of SOD,decreased the content of MDA in cells.In addition,Tau also reduced the intracellular calcium overloading induced by ADR. Conclusion Tau could protect cardiomyocytes from damage induced by ADR through decreasing lipid peroxidation and reversing calcium overloading.
