摘要
目的 观察 90min内血浆异钩藤碱浓度变化对大鼠血压和心脏收缩功能的影响。方法 采用反相高压液相色谱法测定大鼠血浆中异钩藤碱的浓度 ,用RM 6 0 0 0型多导生理记录仪记录大鼠的血压和心脏收缩性能的有关指标。结果 异钩藤碱的有效降压浓度为 0 .38± 0 .0 6~ 2 .36± 0 .44mg/L ,减慢心率及抑制左室压最大变化速率和心肌收缩成分缩短速率等指标的血药浓度为 1.2 7± 0 .0 7~ 2 .36± 0 .44mg/L。 结论 异钩藤碱用于对抗扩血管药引起的加快心率的副作用时 ,血药浓度控制在 1.30mg/L为宜 ;作为降压药使用时 ,其血药浓度最好控制在0 .75mg/L。
Objective effect of the change of isorhynchophylline(IRHY) in plasma on the blood pressure and myocardial contractility of the rats were observed within 90min. Methods The concentration of IRHY in plasma of the rates were determined by a reversed phase high performance liquid chromatographic method, and the tracings of blood pressure and myocardial contractility were recorded on RM 6000 polygraph. Results It was showed that concentration of hypotensive role were about 0.38~2.36mg/L,the concentration of decreasing heart,dP/dTmax and Vmax et al were about 1.27~ 2.36mg/L. Conclusions the results suggested that concentration of IRHY in plasma was controlled about 1.3mg/L was suitable for IRHY used against the side reaction of vasodilator increasing heart rate and suggested that concentration of IRHY in plasma was controlled about 0.75mg/L was suitable for IRHY antihypertensivi.
