外源性GSH镇静催眠作用及其作用机制初探 被引量:1

摘要 目的 观察外源性GSH灌胃给药的镇静催眠作用及对不同脑区NA、5 HIAA和HVA的影响。方法 观察外源性GSH灌胃给药对阈下剂量 (2 5mg/kg ,iv)戊巴比妥钠小鼠行为的影响 ,同时用高效液相色谱电化学法测定了外源性GSH灌胃给药后大鼠不同脑区NA、5 HIAA和HVA的含量。结果 外源性GSH灌胃给药可提高阈下剂量戊巴比妥钠所致的小鼠“睡眠”发生率 ,并使大鼠下丘脑NA下降 ,皮层及脑干 5 HIAA上升。结论 GSH可协同戊巴比妥钠的镇静催眠作用 ,其作用可能与下丘脑NA下降 ,皮层及脑干 5 HIAA上升有关。 Objective To observe the sedation and hypnosis of oral exogenous reduced glutathione(GSH), and the effect on noradre naline(NA), homovanillic acid (HVA), 5 hydroxyindol acetic acid (5 HIAA) contents in different brain areas of rats. Methods The effect of exogenous GSH administered by gavage (i.g) on the sedative and hypnosis of pentobarbital sodium of sub threshold dose(25mg/kg,iv) was observed in mice, and the contents of NA, HVA and 5 HIAA in different brain areas of rats were determined by HPLC with an electro chemical detector. Results The exogenous GSH i.g increased the rate of righting reflex induced by pentobarbital sodium of sub shreshold dose in mice. The contents of 5 HIAA in cortex and brain stem. of rats were increased and the content of NA in hypothalamus of rats was decreased, respectively. Conclusions The exogenous GSH can influence the sedation and hypnosis induced by pentobarbital sodium, which indicated that GSH may be as one of the transmitters in central nerve system, and the changes of 5 HIAA in cortex and brain stem, NA in hypothalamus contribute the synergic effect of GSH to pentobarbital sodium.
