卵巢上皮性癌P170g、GST-π、Top-Ⅱ表达与化疗耐药的关系

EXPRESSIONS OF P170g,GST-π,Top-Ⅱ AND CHEMORESISTANCE IN EPITHELIAL OVARIAN CANCER

目的 :研究卵巢癌组织中P170 g、GST π、Top Ⅱ表达及其与耐药的关系。 方法 :采用免疫组化的方法测定 31例初治、19例术后复发者卵巢癌组织中P170 g、GST π、Top Ⅱ的表达频率。 结果 :①卵巢癌组织中P170 g、GST π、Top Ⅱ表达频率分别为 4 4 % ( 2 /50 )、60 % ( 30 /50 )、4 2 % ( 2 1/50 )。② 31例初治患者卵巢癌组织中三者阳性表达率分别为 35 5% ( 11/31)、4 8 4 % ( 15/31)、54 84 % ( 17/31)。 19例复发患者三者阳性表达率分别为 57 9% ( 11/19)、78 9% ( 15/19)、2 1 1% ( 4 /19)。初治与复发者中GST π、Top Ⅱ表达差异有显著性 (P <0 0 5)。③GST π阳性者化疗有效率 ( 36 67% )明显低于GST π阴性者 ( 70 % ) ,GST π阳性者预后较差。结论 :卵巢癌组织中GST π和Top Ⅱ表达与其化疗耐药有关 ,GST π可作为判断化疗敏感性及预后的因素之一。

Objective:To study the relationship between the expression of p170 glucoprotein(P170 g),glutathione,S transferase π(GST π) and topoisomerase Ⅱ(Top Ⅱ) in operative speciments and chemoresistance in patients with ovarian cancer.Methods:The expression of P170 g,GST π and Top Ⅱ in 50 cases of overian cancer which included 31 initially treated and 19 recurrent patients was retros pectively studied by immunohischemistry.Results:① The positive expression rates of P170g,GST π and Top Ⅱ were 44%(22/50),60%(30/50) and 42%(21/50) respectively.②These expression rates in the 31 initially treated patients were 35.5%(11/31),48 4%(15/31)and 54 8%(17/31),and those of recurrent cases were 57.9%(11/19),78 9%(15/19) and 21.1%(4/19).The expression rates of GST π and Top Ⅱ between the initially treated patients and recurrent cases showed signifficant difference.③Patients with GST π positive expression showed less response to chemotherapy compared with the GST π negative cases,and the GST π positive cases had a poor prognosis.Conclusion:The expression rates of GST π and Top Ⅱ are closely related to the chemoresistance in ovarian cancer.GST π is a useful marker for predicting the chemotherapy sensitivity and the prognosis in the ovarian cancer.
