摘要
目的获得高效价A型肉毒毒素(BoNT/A)表位多肽单克隆抗体。方法利用DNASTAR分析软件,结合Expasy网络服务器,综合参考多参数预测B细胞抗原表位;以多肽作为抗原制备BoNT/AHc表位多肽单克隆抗体,并进行效价鉴定。结果以合成多肽(P1&P2)分别免疫Balb/c小鼠,获得两株单克隆抗体,经鉴定分别是IgG和IgM,效价为10-5~10-6;检测抗体结果显示,纯度较高,特异性好。结论本研究制备出了特异性好、效价较高的A型肉毒毒素单克隆抗体,为获得抗A型肉毒毒素的快速现场检测提供技术支持。
Objective To obtain high - titer monoclonal antibody to botulinum toxin type A (BoNT/A). Methods DNAS- TAR analysis software and Expasy network server were combined to predict the B cell antigenic epitopes, refering to multiple pa- rameters. BoNT/A Hc epitope polypeptide monoclonal antibody was produced based on designed polypeptide, then the titer was evaluated. Results BALB/c mice were immunized by two synthetic peptides (P1 &P2) to obtain two strains of monoclonal an- tibodies ( IgG and IgM), pure and specific, with the titers of 10 -5 - 10-6. Conclusion The high - specificity and high - titer monoclonal antibodies against botulinum toxin A were prepared, thus providing a technical support for the rapid field detection of botulinum toxin type A.
