摘要
目的:建立紫草油中β,β′-二甲基丙烯酰阿卡宁的含量测定方法。方法:采用高效液相色谱(HPLc)法,色谱柱为迪马C18钻石色谱柱(250mm×4.6mm,5μm);流动相为乙腈.水-甲酸(75:25:0.2);检测波长为275nm;柱温为室温。结果:在80.6~564.5ng范围内,β,β′-二甲基丙烯酰阿卡宁进样量和峰面积具有良好的线性关系,线性回归方程为C=1.0279×10^-6A+2.3589×10^-2,r=0.9999。测得紫草油中β,β′-二甲基丙烯酰阿卡宁平均含量为0.8692mg/ml。结论:该方法简便可行,重现性好,可作为紫草油的质量控制方法。
OBJECTIVE:To establish a method for determination of β,β′-dimethylaerylakannin in Shikonin oil. METHODS: The sample was determined by HPLC and separated on chromatographic column Diamonsil C18 (250mm×4.6mm,5μm) with mobile phase consisting of Acetonitril-H2O-HCO2H(75:25:0.2) at detection wavelength of 275 nm. The column temperature was kept at room temperature. RESULTS: The linear range ofβ,β′-dimethylaerylakannin was 80. 6-564. 5 ng, and the linear equation was as follows: C = 1. 027 9 ×10^-6A + 2. 358 9 ×10^-2(r = 0. 999 9). The average content ofβ,β′-dimethylaerylakannin in Shikonin oil was 0. 869 2 mg/ml. CONCLUSION: The method is simple and reproducible and it can be used as the quality control method for Shikonin oil.
