摘要
目的 研究依赖利福平分支杆菌〔1〕(依R菌 )菌株的依R程度、菌落及菌体的形态特点 ,以期为依R菌的定义和鉴定提供依据。方法 从分支杆菌菌株库中选出在含R培养基中生长更旺盛的 2 2株菌株 ,用L -J氏培养基复苏培养。选取单个菌落接种不同R浓度的培养基中培养 ;对不同实验管 (对照管、R管 )中的菌落进行计数、称重、形态观察 ,并作抗酸染色观察菌体形态。结果 2 2株依R菌经菌种鉴定均为结核分支杆菌。依R菌对R的依赖性仅限于某个浓度范围 (R :5 0~6 0 0 μg/ml) ;不同的菌株其依赖范围不同 ,但大都在R10 0~ 30 0 μg/ml范围内生长最旺盛。依R管与非依R管 (对照管 )中菌落(菌体 )形态均有明显的差异 ,其菌落重量差为 2 .2~ 5倍。结论 目前我们所发现的依R菌株均为结核分支杆菌。依R菌对R的依赖为相对依赖。分支杆菌在含R培养基中菌落生长更旺盛和菌体粗大、浓染的特征 。
Aim To study the rifampin-depending degree of rifampin-depending Mycobacterial strains isolated from patients and the characteristics of their colonies and bodies in order to identify the rifampin-depending Mycobacteria in clinic. Methods Twenty two rifampin-depending strains were selected based on their vigorous growth in the media containing rifampin and cultured in the L-J media. The single colony from each culture was inoculated into media containing rifampin (50-800μg/ml); 28 days after that, the color, shape, size and number of colonies were detected, and the colonies were harvested from media and weighted. Results All rifampin-depending strains in this study were identified to be M. tuberculosis. Their dependence of rifampin was limited by rifampin concentration (50~600μg/ml) and most of them grew vigorously in 100~300μg/ml rifampin; the weights of their colonies on the media were 2.2~5 times of controls. Conclusion The rifampin-depending strains identified in our laboratory are M.tuberculosis and their rifampin dependence is relative; the vigorous growth in rifampin-containing media may be a basic characteristic identifying rifampin-depending M.tuberculosis.
