摘要 目的:证明肝细胞生成素(Hepatopoietin,HPO)是否通过自分泌循环的方式刺激体外培养的肝癌细胞系的自主性增殖。方法:以体外培养的肝癌细胞系为模型,利用RTPCR,ELISA和Westernblot观察HPO的表达;利用HPO特异性抗体封闭HPO的作用,观察封闭HPO的作用后,细胞增殖的变化。结果:显示了肝癌细胞表达HPO并可分泌到细胞培养液中,分泌的HPO有刺激肝癌细胞增殖的作用,HPO特异性抗体可中和HPO并可部分抑制肝癌细胞的自主性生长。结论:在体外培养的肝癌细胞中存在维持其自主性生长的HPO自分泌循环。 Objective:The current study was designed to cla rify whether hepatopoietin(HPO)stimulates autonomous growth of hepatoma cell b y autocrine loop.Methods :The authors conducted experiments i n vitro with hepatoma cell lines.RT-PCR,ELISA a nd Western blot were used to examine H PO expression in hepatoma cells.Blocking effect of HPO by HPO neutralizing antibody was utilized and the c hanges of cell proliferation was observed.Results:HPO was expressed by hepatoma cells a nd secreted into the medium.Moreove r,the HPO antibody inhibited specifically the autonomous proliferation of hepatoma cell and antagon ized the stimulatory effect of conce ntrated conditioned medium derived from hep atoma cell HepG2.Conclusion:The results strongly suggest that HPO acts as an autocrine factor to maintain the aut onomous growth of hepatoma cells.
