摘要
目的:建立一种评价肿瘤生物学特性的新方法——p53功能失活检测法并探讨p53功能失活与食管鳞癌TNM分期tumornodesmetastasisstaging和组织学分级的关系。方法:采用p53功能测定法对45例新鲜食管鳞癌组织和正常食管组织进行p53功能检测(p53基因突变检测作为对照)将检测结果与患者的TNM分期和组织学分级进行统计学分析。结果:p53功能失活率为64%,明显高于p53基因突变率49%。p53功能失活和食管鳞癌的TNM分期有关,分期越高,p53功能失活率越高;p53功能失活和食管鳞癌的组织学分级有关,分级越高,p53功能失活率越高。结论:p53功能失活有望成为一种评价食管鳞癌生物学特性的新指标;p53功能失活与食管鳞癌的TNM分期和组织学分级有关。
Objective:The current study was designed to establish a new method to evaluate biolo gical activity of carcinoma —functional status of p53,and investigate the relationship between functional inactivation of p53and the TN M(tumor,nodes,metastasis)staging or histological classifica tion of squamous cell carcinoma of esophagus.Methods :A total of 45samples of fresh esophageal tissues of squamous cell carcinom a and normal esophageal tissues were examined for functional inactivation of p53by detection of functional inactivation of p53(comparison with detection of p53gen e mutation).Then the analyses of detected resul ts and the TNM stagings or the histolo gical classifications of the carcin oma were statistically analyzed in SPSS.Results:The rate of functional inactivation of p53(64%)seemed to be obviously higher than that of p53gene mutation(49%)with a significant difference(P=0.046).There was a significant relationship between functional in activation of p53and the TNM staging of esophageal squamous cell carcinoma.Its rate tended to be increased with the advan ce of the TNM staging;there was a sign ificant relationship between functional inactivation of p53and the histolog ical classification.With the advan ce of the histological classificati on,the rate of functional inactivation of p53coul d have increased.Conclusions :The functional inactivation of p53w ould be expected to be a new method to evaluate the biological activity of esophageal squam ous cell carcinoma ;there was a significant relationship between functional in activation of p53and the TNM staging or the histological classification.
