摘要 通过正交试验和响应面分析法对纳豆激酶液体发酵条件进行了系统研究。由纳豆芽孢杆菌的生长曲线确定出18h^20h为适宜的种龄。采用正交试验法对发酵培养基的组成进行优化,确定出发酵培养基的最佳配方:玉米淀粉2%,动物蛋白胨4%,MgSO4·7H2O0.05%,CaCl20.01%,K2HPO4/KH2PO40.3%/0.1%。通过响应面分析法对发酵条件进行优化,确定出液体发酵最佳条件:初始pH值为6.8,装液量50mL/500mL锥形瓶,接种量3.1%,发酵温度36℃。经过优化,发酵48h后的纳豆激酶酶活从12.47kU/mL提高为32.73kU/mL。 Liquid fermentation conditions of nattokinase production were studied by orthogonal test and response surface analysis(RSA).The growth curve of Bacillus subtilis Natto showed that cultivation for 18h~20h was the best strain age.Orthogonal test was used to optimize the components of the culture medium.Results revealed that the optimum formula of culture medium was as follows:Corn starches 2%,animal peptone 4%,MgSO4·7H2O 0.05%,CaCl2 0.01%,K2HPO4/KH2PO4 0.3%/0.1%.The fermentation conditions were optimized by RSA.It revealed that the optimum liquid fermentation conditions were as follows:initial pH 6.83,volume of liquid 50ml/500ml,inoculation amount 3.14%,fermentation temperature 36.05℃.The optimized conditions allowed nattokinase activity increased from 12.47kU/ml to 32.73kU/ml.
