按照肝脏受 TAA损伤的程度把试验动物 Wistar大白鼠分为两组 ( TAA- 1和 TAA- 2 )。Northern杂交分析表明 ,在 TAA- 1的窦内皮细胞中 ,bcl- x和 bax基因的表达水平轻微降低 ;随着肝损伤的增加 ,表达水平再度下降。核 Run- off转录分析表明 ,两组的窦内皮细胞、总的 RNA合成率下降约 30 %( P<0 .0 0 1 ) ;TAA- 1、bad、bax和 cyclin E的 m RNA合成有所降低 ,而 cyclin A的 m RNA合成则明显增加 ;TAA- 2、bad、bax和 bcl- 2的 m RNA合成增加了 2~ 3倍 ,bcl- x的 m RNA合成增加了
Experimental animals of Wistar rats using oral treatment of Thioacetamide (TAA) were divided into two groups (TAA 1 and TAA 2) according to the extent of liver injury.The analysis by means of Northern blotting showed that a slightly decreased expression level of bcl x and bax was observed in sinusoidal endothelial cells (SEC) of group TAA 1.In SEC of group TAA 2,a further decreased expression of bcl x and bax was more slightly detected.The analysis of nuclear run off transcription assays showed that,in both experimental groups,the rate of incorporation of 32 P UTP into total RNA by isolated nuclei of SEC was significantly reduced by about 30% ( P <0.001) and that the synthesis of mRNA of bad,bax and cyclin E was moderately decreased in SEC of the early phase of liver injury,while that of cyclin A and bcl x was significantly increased.In the late phase of TAA induced fibrogenesis (TAA 2),the synthesis of mRNA of bad,bax and bcl 2 in SEC was increased by 2-3 fold and that of bcl x was raised more than 7 fold.
