摘要
目的探讨依托咪酯对大鼠脑缺血再灌注损伤的保护作用及其机制。方法 SD大鼠60只,随机分为对照组(S)、缺血再灌注组(IR)和依托咪酯组(Etom),各20只。采用4-VO法建立大鼠全脑缺血再灌注模型。缺血再灌注后1d、3d、7d用免疫组化法测定海马CA1区神经元mPTP的表达。结果与S组相比,IR组和Etomidate组海马CA1区神经元mPTP的表达明显上升(P<0.01),但Etom组海马CA1区神经元mPTP的表达低于IR组(P<0.05)。组内各时间点表达水平差异无统计学意义(P>0.05)。结论依托咪酯对大鼠脑缺血再灌注损伤有一定的保护作用,可能是通过抑制mPTP的表达而发挥作用的。
Objective To investigate the protective effects and possible mechanisms of Et0midate against global brain is. chemia-reperfusion injury in SD rats. Methods Twenty-seven SD rats were randomly divided into three groups: sham operation group(S) ; IR group and Etomidate(Etom) group. Global brain ischemic model was established by 4-VO method. The expression of mPTP was determined by immunochemistry the 1 st day, 3th day, 7 th day after global brain ischemia-reperfusion injury. Results Compared with sham group, the expression of mPTP was significantly increased in IR group and Etom-group (P 〈 0. 01 ) ; but the expression of mPTP in Etom-group than IR group. Conclusion Peritonael injection of Etomidate before ischemia could protect the brain against IR injury in SD rats through an inhibition of mPTP expression.
