摘要
利用 SSP- PCR技术对 S组 CMS与正常胞质线粒体胞质育性相关基因 cox ,cox 的 5′上游、 3′下游以及基因编码区与正常 N胞质进行了遗传多型性的比较研究。结果表明在所选用的 (特异引物 +随机引物 )的引物组合中 ,未能在 cox 基因的 5′上游和 3′下游区发现变异 ,但其基因内含子中存在基于 SSP- PCR的遗传多型性。在 S组不育系的 cox 基因 5′上游调控区内 ,由于复杂的重组事件而导致翻译起始密码 ATG和富含 AT的核糖体结合区域的缺失。讨论了 SSP- PCR技术作为揭示和分析某一目的基因的遗传变异的有效性与准确性 ,以及 cox 基因在表达水平上的变异可能与玉米
The PCR technique with single special primer combine with random primer (SSP PC R) were used to research the genetic heterogeneity of cox[KG-4]Ⅰ, cox [KG-4]Ⅱ among S CMS line and normal fertile line in maize. The results are as follows; No divers ity were found on the 5′ upstream and 3′ downsream of the cox[KG-4]Ⅱ gene, but a di fferent map pattem of SSP PCR have been detected on the intron of cox[KG- 4]Ⅱ ; A deletion of ATG site of initiation codon and a ribosome binding region which ric h AT were found at the 5′ upstream of cox[KG-4]Ⅰ in S CMS, which suggest that t he dropping down at the transcriptional level of mRNA maybe correlated with the cytoplasmic male sterility. The SSP PCR is suitable for research the genetic he terogeneity and insertion sites of transposon.
