过表达Tribbles同源蛋白3促进成纤维细胞分泌I型胶原的研究 被引量:3

摘要 目的探讨过表达Tribbles同源蛋白3对小鼠成纤维细胞3T3-NIH分泌胶原的影响。方法使用腺病毒载体过量表达Tribbles同源蛋白3,使用PCR以及Western blot方法检测Tribbles同源蛋白3的过量表达情况,使用天狼猩红染色试剂盒以及Western blot检测胶原的表达。结果使用过量表达Tribbles同源蛋白3的腺病毒载体可以增加成纤维细胞3T3-NIH中Tribbles同源蛋白3的转录水平以及蛋白水平的表达,并增加3T3-NIH细胞中I型胶原的蛋白表达水平,降低III型胶原的蛋白表达水平。结论过量表达Tribbles同源蛋白3可以促进小鼠成纤维细胞I型胶原的表达与分泌。 Objective The aim of this study is to investigate the effect on secretion of collagen with overexpression of tribbles homolog 3 (TRB3) in mouse fibroblast 3T3-NIH. Methods Using adenoviral vectors to overexpress TRB3, and the expression of TRB3 was detected by PCR and Western blot. The expression of collagen was detected by Sirius red staining kit and Western blot. Results The expression of TRB3 was significantly increased after infecting adenoviral vector. After overexpress TRB3 specifically, the expression and secretion of type I collagen were up-regulated in the mouse fibroblast cells, but the expression of type III collagen was decreased. Conclusion The overexpression of TRB3 promotes the secretion and expression of type I collagen in mouse fibroblasts cells
