摘要
目的:探讨柴胡皂甙D对肺癌A549细胞的放射增敏作用及机制。方法:利用细胞克隆技术进行剂量-存活曲线分析,MTT法检测细胞凋亡,流式细胞仪检测细胞周期,并对细胞周期进行分析。结果:柴胡皂甙D组D0值为2.7Gy,空白对照组D0值为3.6Gy(P<0.05)。随着柴胡皂甙D浓度的增加及作用时间的延长肺癌A549细胞抑制率增加(P<0.05),且经柴胡皂甙D处理后,与对照组相比,S期细胞比例降低,G2/M期细胞比例增高。结论:柴胡皂甙D可以明显提高肺癌A549细胞的放射敏感性,机制可能与其引起瘤细胞周期阻滞,诱导肿瘤细胞凋亡有关。
Objective:To investigate the influence of SSD on the radiosensitivity of lung cancer cell lines (A549) and its mechanism. Methods:The colony formation ability of A549 cells after X - ray irradiation was analyzed by linear quadratic formulation. The apoptotic percentage was measured by MTT and cell cycle was analyzed by FCM. Resuits : The Do value of A549 cell lines was 2.7 Gy, and that of control group was 3.6Gy, respectively (P 〈 0.05). The apoptotic percentage of A549 cell lines was eulaged with the increased of the concentration of SSD (P 〈 0.05). in SSD group the percentage of cell sin G2/M phase increased. Conclusion: SSD cart increase the radiosensitivity of lung cancer cell lines,which may be associated with blocking cell cycle and inducing apoptosis of A549 ceils.
