Chiral Resolution of Propisochlor Enantiomers by High Performance Liquid Chromatography

摘要 本研究旨在建立一种快速高效的异丙草胺对映体色谱拆分方法.首次筛选比较了9种涂覆型或键合型纤维素类和直链淀粉类不同手性固定相对异丙草胺对映体的拆分性能,系统考察了醇改性剂种类、比例、流动相流速、柱温等因素对拆分效果的影响,确立在Chiralpak AD-H色谱柱上,以正己烷/异丙醇(V/V,99/1)为流动相,柱温35℃,流速1.0mL/min,异丙草胺对映体可得到最优拆分,分离度为2.41.对异丙草胺对映体在手性固定相上保留和分离的热力学机理进行了初步探讨,发现其手性拆分过程受焓控制. This investigation aims to establish a fast and efficient chromatographic resolution for the axially chiral herbicide propisochlor. Firstly, the properties of 9 kinds of chiral stationary phases coated or chemically bonded cellu- lose and amylose type were compared. The effects of alcohols modifiers, mobile phase ratio, flow rate and temperature on the chromatographic performance were systematically investigated. The best resolution of propisochlor on Chiral- pak AD-H was obtained with the mobile phase of n-hexane/iso-propanol (V/V,99/1), at a flow rate of 1.0 mL/min and 35℃, and the resolution could reach 2.41. The thermodynamical mechanism of enantioseparation had also been preliminarily studied and the enantioseparation was controlled by enthalpy.
