肝细胞核因子-1α对小鼠Ⅰ型葡萄糖载体表达的影响

摘要 [目的]探讨在CV-1细胞中肝细胞核因子-1α(HNF-1α)对小鼠Ⅰ型葡萄糖载体(mGLUT1)表达的影响.[方法]利用DNA克隆技术,将HNF-1αcDNA序列重组到表达载体pSV-sport上,以HNF-1α转染CV-1细胞,应用Northern blot方法测定HNF-1α对mGLUT1 mRNA表达的影响.[结果]HNF-1α可提高mGLUT1 mRNA表达水平及稳定性.[结论]HNF-1α可增强mGLUT1的表达,可能通过调节GLUT1的表达参与胰岛素对血糖的调节过程. OBJECTIVE To study the effect of hepatocyte nuclear factor lu (HNF-1α) on expression of mouse glucose transporter I (mGLUT1) in CV-1 cells. METHODS Using DNA cloning technology, HNF-1α cDNA sequence recombined into pSV-sport expression vector, and the recombinant HNF-1α was transfected into CV-1 cells, and the effect of HNF-1α on expression of mGLUT1 mRNA was detected by Northern blot. RESULTS HNF-1α can increase the expression and stability of mGLUT1 mRNA. CONCLUSION HNF-1α increases expression of mGLUT1, and may participate in regulating process of insulin for blood glucose by regulating the expression of mGLUT1.
