丹参酮ⅡA对阿尔茨海默病模型大鼠脑组织caspase-3、Akt与NF-κB表达的影响 被引量:15

Effects of tanshinone ⅡA on the expressions of caspase-3,Akt and NF-κB in the brains of rat models of Alzheimer's disease

摘要 目的观察丹参酮ⅡA(TanⅡA)对β淀粉样蛋白(Aβ)诱导的大鼠脑组织Akt、NF-κB、caspase-3表达水平的影响。方法 30只雄性SD大鼠随机分为假手术组、AD模型组和TanⅡA治疗组。在大鼠脑海马内注射Aβ方法建立AD动物模型。免疫荧光法检测Aβ1-16标记部位和表达水平,免疫组化法检测caspase-3的表达水平,免疫组化染色和Western blot法检测Akt、NF-κB的表达水平。结果 Aβ1-16在假手术组中无表达,而在AD模型组和TanⅡA治疗组二者间Aβ1-16表达水平无明显差异(P>0.05)。Akt的表达在模型组低于假手术组和TanⅡA治疗组,差异有统计学意义(P<0.05),而模型组NF-κB与caspase-3的表达水平明显高于假手术组和TanⅡA组,差异显著(P<0.05)。结论 TanⅡA可降低模型鼠NF-κB与caspase-3的表达水平,且上调Akt的表达。 Objective To observe the effects of tanshinone II A (Tan II A) on the expressions of serine/threonine kinase Akt, nuclear factor KB (NF-KB) and caspase-3 in the brain tissues of rat models of Alzheimer's disease (AD). Me,otis Thirty male Sprague-Dawley rats were randomized into three groups ( n = 10 per group) : Sham group, AD group, Tan I1 A treatment group. The models of AD were established by injecting 15-amyloid (AI3) into the hippocampus of rats. The location and expression level of Aβ1-16 was observed by immunofluorescence, the expression level of caspase-3 was detected by immuno- histochemistry, and the expression levels of Akt and NF-KB were determined by immunohistochemistry and Western blotting. Results There was no Aβ1-16 detected in the control group, and the difference in the expression level of Aβ1-16 between AD group and Tan I1 A treatment group was not significant statistically ( P 〉 0.05) ; The expression of Akt in AD group was lower than that in the control group and the Tan II A treatment group, and the difference between AD group and the other two groups was of statistical significance ( P 〈 0.0.5). Conversely, compared with Tan II A treatment group and sham group, the AD group was significantly higher in the expression levels of NF-kB and caspase-3 (P〈0.05). Conclusion Tan I1 A can up-regulate the expression of Akt and inhibit the production of NF-KB and casloase-3 in the model rats with AD.
