Th1型细胞因子在胶原蛋白诱导性关节炎小鼠血清中的动态变化 被引量:15

摘要 目的研究鸡Ⅱ型胶原蛋白诱导性关节炎(CIA)小鼠血清Th1型细胞因子IFN-γ、IL-2、TNF-α的动态变化。方法 72只DBA1/J小鼠随机分为正常对照组(每组6只)和模型组(每组12只),分别在加强免疫后第7、14、21、35天,无菌摘眼球取血清,应用流式细胞术检测各组小鼠血清中细胞因子IFN-γ、IL-2、TNF-α在关节炎不同时期的动态变化情况。结果模型组IFN-γ在加强免疫后第7天和第14天显著升高(P<0.05),加强免疫后第21天、第35天开始下降,与正常组比较无区别(P>0.05);模型组IL-2和TNF-α在加强免疫后第7天上升明显,与正常组比较有统计学意义(P<0.05),加强免疫后第14、21、35天IL-2和TNF-α逐渐下降,与正常组比较无统计学意义(P>0.05)。结论 CD4+Th1型细胞因子参与CIA发病全过程,而且在不同的发病阶段其相关细胞因子的变化与关节炎的炎症进展有一定关系。 Objective To study the dynamic changes of Thi-type cytokines IFN-y, IL-2, TNF-α in the serum of chicken type Ⅱ collagen-induced arthritis (CIA) in mice. Methods A total of 72 DBA1/J mice were randomly divided into control group (n =24) and model group (n =48), and each group was subgrouped into 4 groups (n=6 in control subgroup and n = 12 in model subgroup) according to the time of taking the eyeball for blood and separating the serum under sterile condition (7, 14, 21 and 35 days after booster immunization). The dynamic changes of cytokines IFN-γ, IL-2 and TNF-α in different periods were detected by flow cytometry. Results The level of IFN-y in the model group was significantly higher than that in the control group on the 7 day and 14 day after booster immunization ( P〈0.05), and it began to decline on the 21 day and there were no significant differences between the model group and the control group on the 21 day and 35 day ( P 〉 0.05). The levels of IL-2 and TNF-α in the model group on the 7 day after booster immunization significantly increased as compared with those of the control group ( P 〈 0.05), however, on the 14, 21 and 35 day there were no significant differences between the model group and the control group (P 〉0.05). Conclusion CD4 + Th1 cytokines participate in the pathogenesis of CIA and their alterations at different stages of the disease have a relation to the development of arthritis.
