摘要
目的 探讨脐血与健康供者稳态骨髓中血管内皮生长因子(VEGF)、转化生长因子β1(TGF-β1)、Tie-2、血管生成素(Ang-1、Ang-2)的血浆浓度变化.方法 采用酶联免疫吸附测定(ELISA)方法对32例脐血和健康供者稳态骨髓的VEGF、TGF-β1、Tie-2、Ang-1及Ang-2的血浆浓度进行检测.结果 脐血中VEGF的水平与稳态骨髓比较显著升高446.62(44.31-1 430.46) ng/L vs 131.62(16.62-1 201.92) ng/L(P〈0.01).脐血中TGF-β1的水平与稳态骨髓比较却显著减低276.36(80.91-880.91) ng/L vs 476.36(235.45-1 390.00) ng/L(P<0.01).稳态骨髓中Tie-2的血浆浓度与脐血比较差异无统计学意义(P>0.05).尽管骨髓中Ang-1、Ang-2的浓度与脐血相比,有增加的趋势,但是差异均无统计学意义(P>0.05).结论 脐血和稳态骨髓中VEGF和TGF-β1水平的差异提示脐血和骨髓造血干细胞所处的微环境可能不同.
Objective To explore the alteration of vascular endothelial growth factor (VEGF), transforming growth factor-β1 (TGF-β1),Tie-2 ,angiopoietins(Ang-1 ,Ang-2) in umbilical cord blood(UCB) and bone marrow(BM). Methods Using ELISA method,the plasma levels of TGF-β1, VEGF,Tie-2, Ang-1 and Ang-2 were detected in 32 UCB and BM donors. Results The plasma level of VEGF in UCB was significantly higher than in BM, 446.62 (44.31- 1 430.46) ng/L vs 131.62(16.62-1 201.92) ng/L( P〈0.01). The plasma level of TGF-131 in UCB was significantly lower than in BM,276.36(80.91-880.91)ng/L vs 476.36(235.45-1 390.00)ng/L( P 〈0.01). The median Tie-2,Ang- 1 and Ang-2 levels did not differ between UCB and BM samples ( P 〉0.05). Conclusion The differences in VEGF and TGF-β1 suggest deviations in placenta and BM microenvironment.
