重组人球状脂联素抑制MCF-7细胞生长及NF-κB的相关性 被引量:2

摘要 目的探讨重组人球状脂联素是否通过核因子κB(NF-κB)通路对MCF-7细胞的增殖、凋亡产生影响。方法不同浓度(0、10、20、40、80 ng/ml)重组人球状脂联素培养MCF-7细胞24、48 h。NF-κB活化抑制剂二硫代氨基甲酸吡咯烷(PDTC)预处理细胞1 h后,不同浓度重组人球状脂联素再作用MCF-7细胞24 h,采用四甲基偶氮唑蓝(MTT)法检测MCF-7细胞增殖活性;流式细胞仪(Annexin-FITC/PI双染)检测细胞凋亡率;Western blot法测定NF-κB蛋白表达。结果与对照组相比,40、80 ng/ml重组人球状脂联素作用细胞后,MCF-7细胞生长受到抑制(P<0.05),NF-κB蛋白表达量减少(P<0.05),抑制效应均呈时间-剂量依赖性;PDTC可以阻断重组人球状脂联素对MCF-7细胞活性抑制及NF-κB蛋白表达抑制作用;80 ng/ml重组人球状脂联素作用细胞24 h,总凋亡率为(13.180 0±1.064 9)%,与对照组相比差异无统计学意义。结论重组人球状脂联素可以抑制MCF-7细胞生长,效应呈时间-剂量依赖性,其机制可能与NF-κB通路相关。 Objective To explore whether there is the effect of the globular adiponectin on MCF-7 cells prolitora- lion and apoptosis via nuclear factor kappa B (NF-KB) pathway. Methods Developing breast cancer ceils MCF-7 in the high sugar environment. The cells were treated with different concentrations of globular adiponectin (0,10, 20,40,80 ng/ml) for 24 h and 48 h, different concentrations globular adiponectin treated cells for 24 h again after pretreatment of NF-KB inhibitors PDTC (50 retool/L). The cell survivals were determined by MTT method. The flow cytometry with Annexin-FITC/PI double staining detected apoptosis. Western blot detected the expression of NF-κB protein. Results Compared with NC group, MCF-7 cells viability and the expression of NF-KB protein were decreased by increasing concentrations and time of globular adiponectin, especially for the concentration of 40 and 80 ng/ml(P 〈 0. 05). Cells used by pretreatment of PDTC, the effect of globular adiponeetin for cells growth and the NF-κB proterin were blocked.
