The expression of Kindlin-2 in hepatocellular carcinoma and its relationship with tumor angiogenesis

摘要 目的探讨黏着斑蛋白(Kindlin-2)在肝细胞癌(简称肝癌)中表达的临床意义及其与肝癌血管生成的关系。方法采用逆转录-聚合酶链反应(RT-PCR)方法检测12例原发性肝癌及癌旁配对组织和6例正常肝组织中Kindlin-2基因的表达水平。采用免疫组化方法检测69例原发性肝癌患者癌组织及癌旁组织中Kindlin-2蛋白的表达情况,以及肝癌组织中血管内皮生长因子(VEGF)和CD34的表达量。结果Kindlin-2在肝癌组织中的表达明显高于癌旁及正常肝组织;Kindlin-2的表达与肿瘤直径(P=0.010)、门静脉侵犯(P=0.025)相关;肝癌组织中Kindlin-2表达与VEGF表达呈正相关(r=0.401,P=0.001),且Kindlin-2阳性者微血管密度(MVD)值高于阴性者,差异有统计学意义(P<0.01)。结论肝癌组织中异常表达增高的Kindlin-2与肿瘤微血管生呈正相关,提示Kindlin-2可能成为肝癌抗血管治疗的潜在靶点。 Objective To explore the clinical significance of Kindlin-2 expression in hepatocellular carcinoma ( HCC) and its relationship with HCC angiogenesis. Methods RT-PCR was performed to detect the gene expres-sion of Kindlin-2 in 12 cases of primary HCC tissues and adjacent tissues, and 6 cases of normal liver tissues. Im-munohistochemistry was used to determine the protein expression of Kindlin-2 in HCC and adjacent tissues from 69 primary HCC patients. Furthermore, vascular endothelial growth factor ( VEGF) and CD34 were also examined in HCC tissues mentioned above. Results Kindlin-2 expression appeared at a high level compared to adjacent tissues and normal liver tissues. The expression of Kindlin-2 was associated with tumor diameter ( P=0.010 ) and portal venous invasion ( P=0.025 ) . The expression of Kindlin-2 in HCC was positively correlated with VEGF expression (r=0.401, P=0.001);besides, tumors with Kindlin-2 positive expression exhibited higher MVD than that nega-tive expression tumors(P&lt;0.01). Conclusion Aberrant overexpression of Kindlin-2 in HCC is closely associated with tumor angiogenesis, and Kindlin-2 can be the potential targets of anti-angiogenesis therapy of HCC.
