前列腺素E_2对新生大鼠呼吸影响的机制研究

摘要 目的 探讨SD新生大鼠右心房注射前列腺素E2 (PGE2)产生呼吸暂停的作用机制.方法 将24只7~9天的SD大鼠随机分为3组,分别在右心房注射辣椒平、S-3144(P物质受体拮抗剂)和AH6809(前列腺素E2受体拮抗剂)前后注射PGE2,观察呼吸指标的变化.结果 右心房注射PGE2在7~9天大鼠引起呼吸暂停;AH6809可缩短PGE2产生的呼气时间约42% (P<0.05),但辣椒平和S-3144对PGE2产生的呼吸暂停无明显影响.结论 新生大鼠右心房注射PGE2引起呼吸暂停,是通过作用于前列腺素E2(EP2)受体产生而与辣椒素受体和P物质受体无关. Objective To explore the possible mechanism of right atrial injection of prostaglandin E2 (PGE2) on respiratory response in neonatal Sprague Dawley rats. Methods 24 SD rats at postnatal day 7-9( P7- 9) received bo- lus injection of PGE2 before and after capsazepine, S-a144 (antagonist of NK1 receptor), and AH6809 (antagonist of EP2 receptor) injection from the right atrium. The ventilatory alterations were observed and analyzed. Results Right at- rial bolus injection of PGEz generated apnea in PT 9 rats. AH6809 could evidently reduce the duration of expiration in- duced by PGE2 approximately 42~ (P〈0.05), but capsazepine and S-3144 could not change. Conclusion Apnea induced by the right atrial injection of PGFe is partially induced by activating EP2 receptor but not capsaicin and neurokinin 1 receptors.
