“双固一通”电针对老年阳虚模型大鼠淋巴细胞凋亡和促炎因子IL-6、IL-1的影响 被引量:8

摘要 目的探讨"双固一通"电针治疗对老年阳虚模型大鼠脾脏淋巴细胞凋亡和血清促炎因子IL-6及IL-1的影响。方法选用SD大鼠,随机分为正常对照组(NC)、老年阳虚模型组(Model)、"双固一通"电针组(EA)和电针对照组(EAc)。除NC组外,其余3组大鼠均通过皮下注射D-半乳糖40 d,后继续肌肉注射氢化可的松7 d,诱导为老年阳虚模型。EA组取关元、后三里和百会穴治疗,EAc组取中极、阴陵泉、印堂穴治疗。每星期治疗6次,共治疗4星期。采用Annexin V双染流式细胞术检测大鼠脾脏淋巴细胞凋亡率;酶联免疫吸附法检测血清中IL-6、IL-1含量的变化。结果与NC组相比,Model组大鼠脾脏淋巴细胞凋亡率显著上升(P<0.01),血清IL-6及IL-1含量均显著增高(P<0.01);与Model组相比,EA组和EAc组大鼠上述指标均明显降低(P<0.01,P<0.05),且EA组上述指标下降更显著,与EAc组比较,差异具有统计学意义(P<0.05)。结论电针能明显降低老年阳虚模型大鼠脾脏淋巴细胞凋亡率和血清中促炎因子IL-6、IL-1的含量,提示电针对老年阳虚证导致的免疫失衡和炎性因子失衡有治疗作用,且"双固一通"配穴的电针疗效优于邻近对照配穴的疗效。 Objective To investigate the effect of "double reinforcing and one unblocking" electroacupuncture on splenic lymphocyte apoptosis and serum inflammatory factors IL-6 and IL-1β in a rat model of senile yang deficiency. Method SD rats were selected and randomly allocated to normal control group (NC), senile yang deficiency model group (model), "double reinforcing and one unblocking" electroacupuncture group (EA) and electroacupuncture control group (EAc). A model of senile yang deficiency was made by 7 days' intramuscular injection of hydrocortisone following 40 days' subdermal injection of D-galactose in all the groups of rats except the NC group. Points Guangyuan, Housanli and Baihui were selected in the EA group and points Zhongji, Yinlingquan and Yintang, in the EAc group. Treatment was given 6 times a week for a total of 4 weeks. Rat splenic lymphocyte apoptosis rate was determined using Annexin V double staining flow cytometry. Serum IL-6 and IL-1β contents were measured by enzyme-linked immunoadsorbent assay. Result Splenic lymphocyte apoptosis rate and serum IL-6 and IL-1βcontents increased significantly in the model group of rats compared with the NC group (both P〈0.01). The above indices decreased significantly in the EA and EAc groups of rats compared with the model group (P〈0.01, P〈0.05). The above indices decreased more in the EA group than in the EAc group; there was a statistically significant difference (P〈0.05). Conclusion Electroacupuncture can significantly decrease splenic lymphocyte apoptosis and serum inflammatory factors IL-6 and IL-1βcontents in a rat model of senile yang deficiency, suggesting that electroacupuncture has a therapeutic effect on immune imbalance and inflammatory factor imbalance due to senile yang deficiency. The therapeutic effect of electroacupuncture by "double reinforcing and one unblocking" point combination is better than that of electroacupuncture by control adjacent point combination.
