同步衍生化法测定龙葵中还原型谷胱甘肽(GSH)和总巯基(—SH)含量 被引量:1

Determination of Reduced Glutathione( GSH) and Total Thiols( —SH) in Solanum Nigrmn L. by in Situ Derivatization Methods

摘要 用衍生化试剂邻苯二甲醛(OPA)和4,4'-二硫代联吡啶(4-DPS),采用细胞破壁、衍生化为一体的同步提取方法,分别采用荧光光度法和UV-Vis分光光度法测定了龙葵鲜叶中还原型谷胱甘肽(GSH)和总巯基(—SH)含量。龙葵鲜叶GSH和总巯基(—SH)含量分别为0.115 1和0.387 8μmol/g,方法的回收率分别达101.78%和98.36%,检测限分别为0.154 7和0.899 4μmol/L。该方法灵敏、可靠且满足植物鲜品巯基化合物的灵敏测定。 Reduced glutathione and total thiols have been de- termined in the fresh leaves of Solanum nigrmn L by in situ derivatization during ultrasonic cell rupture using o-phthalde- hyde ( OPA ) and 4,4'-dithiodipyridine (4-DPS) as derivat- izing agents by fluorophotometric and UV-Vis spectrophoto- metric methods. The amounts of GSH and total thiols (-SH) determined in the fresh leaves of Solanum nigrmn L are 0. 115 1μmoL/g and 0. 387 8μmoL/g respectively. The recoveries of the methods are 101.78% and 98.36% , detection limits are 0. 154 7 μmol/L and 0. 899 g μmol/L respectively. The methods are sensitive and accurate for the determination of thiol compounds in fresh plant samples.
