溶血性链球菌制剂OK-432对二甲基苯丙蒽/巴豆油诱导小鼠皮肤乳头状瘤的预防作用

摘要 目的构建二甲基苯丙蒽(DMBA)/巴豆油小鼠皮肤乳头状瘤模型,应用其研究溶血性链球菌制剂OK-432对化学致癌剂的对抗作用。方法雌性DBA/2小鼠40只,随机分为模型组和OK-432组。设计经典的DMBA/巴豆油诱导小鼠皮肤乳头状瘤模型,从肿瘤发生率、每鼠荷瘤数、抑瘤率、肿瘤体积变化和皮肤组织病理切片5方面对2组小鼠进行药效评价,探讨OK-432对皮肤乳头状瘤生成的抑制作用。结果 OK-432组比模型组肿瘤发生时间推迟近2.5周,肿瘤诱生率和每鼠荷瘤数减小,抑瘤率为65.6%,皮肤组织病理切片也有显著差异。结论 OK-432能在一定程度上推迟乳头状瘤的发生时间,减少肿瘤的发生,OK-432有一定的癌症化学预防作用。 Objective To construct the 7, 12- dimethylbenz anthracene (DMBA)/croton oil-induced skin papil- loma model in mice, and to investigate the effects of anti-tumorigenesis capacity of OK-432 when subject to chemical carcinogens. Methods Forty female DBA/2 mice were randomly allocated to model group and OK-432 group. The mouse skin papilloma model induced by DMBA/croton oil was established, which entailed the comparison on the inci- dence of tumor, the count of papilloma, tumor inhibitory rate, changes in the volume of tumor and tissue pathological section. Results The OK-432 group resulted in a lag of 2.5 weeks of tumorigenesis compared with the model group. The OK-432 group was associated with reduced incidence of tumorigenesis and the number of tumors, resulting in an inhibitory rate of 65.6%. The difference in histological examinations of skin sections was also remarkable (P〈0.05). Conclusion OK-432 results in, to a certain extent, a time lag in the incidence of tumorigenesis and reduces the inci- dence rate of tumors and therefore should be regarded as an effective chemopreventive agent for cancer.
