多巴酚丁胺对常见上皮源性肿瘤细胞增殖的影响

摘要 目的观察多巴酚丁胺对多种常见上皮源性肿瘤细胞株的增殖抑制作用。方法收集不同浓度多巴酚丁胺处理的胃癌SGC-7901细胞、结肠癌HT-29细胞、卵巢癌HO-8910细胞、宫颈癌SiHa细胞和肺癌A549细胞,于24 h、48 h和72 h采用噻唑蓝(MTT)法检测抑制率,流式细胞仪检测凋亡率。结果不同浓度的药物处理24 h、48 h和72 h后对以上5种上皮源性肿瘤细胞均有一定的抑制作用,随着药物浓度的增加和时间的延长,抑制率有不同程度的增加。30μmol/L药物处理72 h之后对各组细胞的抑制作用最强,由强到弱依次为SGC-7901(41.57±1.10)%、A549(24.32±2.22)%、SiHa(20.66±2.60)%、HT-29(20.38±1.60)%和HO-8910(15.25±0.73)%,对SGC-7901的抑制作用较强,与其他几种细胞差异有统计学意义(P<0.05)。该浓度在72 h对各肿瘤细胞的凋亡率以胃癌SGC-7901和肺癌A549细胞较明显,与对照组细胞差异有统计学意义(P<0.05)。结论多巴酚丁胺对上皮源性肿瘤细胞有一定的增殖抑制作用,并且呈浓度和时间依赖性,该药可明显抑制胃癌细胞增殖,并诱导其凋亡,具有潜在应用价值。 Objective To investigate the inhibitory effects of dobutamine on common epithelial tumor cells proliferation. Methods Different tumor cell lines, including gastric carcinoma SGC-7901 cells, colorectal carcinoma HT-29 cells, ovarian carcinoma H0-8910 cells, cervical carcinoma SiHa cells and lung cancer A549 cells, were incu- bated with different concentrations of dobutamine. This entailed detection of the proliferation inhibition rate by MTr as well as apoptosis by flow cytometer at 24 h, 48 h and 72 h, respectively. Results Dobutamine exhibited a certain inhibitory effects, in a concentration-dependent and time-dependent fashion, on the proliferation to these types of ep- ithelial tumors cells at 24 h, 48 h and 72 h. Incubation with 30 umol/L of dobutamine for 72 h led to the most re- markable inhibitory effect on the proliferation inhibition rate [(41.57±1.10)% for SGC-7901 cells, (24.32±2.22)% for A549 cells, (20.66±2.60)% for SiHa cells, (20.38±1.60)% for HT-29 cells and (15.25±0.73)% for H0-8910 cells] (P〈 0.05). Incubation with 30μmol/L dobutamine at 72 h yielded the most marked apoptotic rate of SGC-7901 and A549 cells compared with control group. (P〈0.05). Conclusion Dobutamine inhibits the proliferation and induces the apoptosis of epithelial tumors cells, in particular the gastric carcinoma cells, in a concentration-dependent and time- dependent manner and demonstrates the potential significance for future application.
