摘要
目的通过检测再生障碍性贫血(AA)患者骨髓单个核细胞表面肿瘤坏死因子相关凋亡诱导配体(TRAIL)的表达,探讨TRAIL在AA造血干细胞凋亡中的作用。方法采用流式细胞术对12例AA患者(AA组)、11例AA治疗后缓解患者(缓解组)、12例正常对照者(对照组)骨髓单个核细胞表面TRAIL的表达进行检测。结果 AA组、缓解组、对照组TRAIL均有表达;AA组TRAIL的阳性表达率(5.8±3.3)%高于缓解组(2.4±2.0)%和对照组(3.2±2.2)%(P<0.05);缓解组与对照组间差异无统计学意义(P>0.05)。结论 TRAIL在AA患者骨髓单个核细胞表面的表达阳性率增高,提示TRAIL介导的细胞凋亡可能在AA的发病机制中发挥一定的作用。
Objective- To detect the expression of tumor necrosis factor-related apoptosis-inducing ligand @RAIL) on the surface of mononuclear cells in patients with aplastic anemia (AA), and to investigate the effects of TRAIL on the apoptosis of hemopoietic stem cells. Methods The flow cytometry was employed to detect the expres- sion of TRAIL on the bone marrow mononuelear cells in 12 patients with AA (AA group), 11 patients with remission of AA (remission group) and 12 normal subjects (control group). Results TRAIL was expressed in these three groups. The level of TRAIL expression in AA group (5.8±3.3)% was markedly higher than that in remission group (2.4±2.0)% and control group (3.2±2.2)% (P〈0.05), with no significant difference being shown between the latter two groups (P〉 0.05). Conclusion The expression of TRAIL is augmented on the surface of bone marrow mononuclear cells, suggest- ing that the associated apoptosis may have played a role in the pathogenesis of AA.
