摘要
研究旨在建立一种检测饲料中AC4含量的高效液相色谱方法。饲料经乙酸乙酯提取,正己烷液液萃取净化,以甲醇-0.2%磷酸水溶液(体积比65:35)为流动相,在C18色谱柱上,紫外检测器(波长251nm)下分离检测。该方法有很好的线性关系,在添加浓度范围内,AC4的平均回收率为98.4%-106.4%,日内变异系数为0.7%~4.6%,日间变异系数为3.6%~4.3%经验证该方法操作简单,灵敏度高,准确性和重现性良好,可用于测定饲料中AC4含量的测定。
This study aimed to establish a kind of HPLC method for detecting AC4 in feed. Extraction used ethyl acetate and was cleaned up with n-hexane, and AC4 was separated on C18 column with UV detector at 251 nm, using methanol and 0.2% orthnphosphorie acid buffer (V: V=65:35) as mobile phase. This method has a good linear relationship. The average reeovery of AC4 ranged from 98.4%-106.4%, with RSD of 0.7%-4.6% within group, and 3.6%-4.3% between group. The methnd was test-ed to be of convenient, high sensitivity, good accuracy and precision.
