不同种类药物对肿瘤细胞增殖的抑制作用 被引量:2

Correlation between glioma cells chemotherapy drugs sensitivity tests in vitro and molecular features

摘要 目的 对胶质瘤肿瘤细胞进行原代培养及体外药物敏感实验,检测不同药物及药物浓度对具备各型分子生物学特征的肿瘤细胞增殖的抑制作用.方法 取38例各病理类型的胶质瘤新鲜肿瘤标本,进行体外原代肿瘤细胞培养.在培养期间将肿瘤细胞进行分组并加入各种浓度的卡铂、福莫司汀、替莫唑胺、替尼泊苷和长春新碱,采用噻唑蓝(MTT)比色法和流式细胞术检测化疗后的细胞活性,对结果进行统计.结果 卡铂、福莫司汀、替莫唑胺、替尼泊苷和长春新碱均表现出了不同程度的抗肿瘤作用,对38例胶质瘤细胞的作用敏感率依次为26.32%、34.21%、44.74%、57.89%、15.79%.烷化剂类药物对O6甲基鸟嘌呤-DNA甲基转移酶表达阳性的胶质瘤和表达阴性的胶质瘤之间的作用差异有统计学意义(P<0.01).高级别胶质瘤的耐药现象比低级别胶质瘤更为显著.结论 不同类型的化学治疗药物对胶质瘤细胞的抑制效果不尽相同,Ⅳ级胶质瘤的耐药性较其他级别者更为明显,O6甲基鸟嘌呤-DNA甲基转移酶的表达会使肿瘤细胞对烷化剂类化疗药物产生一定的耐受.采用流式细胞术检测获得的实验结果敏感度高于MTT,但特异度降低. Objective To explant culture primary glioma cells and test chemotherapy drugs sensitivity in vitro,study the differences of drug types and concentrations in the inhibition effect on cells of distinct molecular features.Methods 38 glioma cells diagnosed as glioma surgical removal patients and grouped in random and cultured with carboplatin,fotemustine,temozolomide,teniposide and vincristine,cells activity were tested by methyl thiazol tetrazolium (MTT) assay and flow cytometry.Results Carboplatin,fotemustine,temozolomide teniposide and vincristine were all exhibited the different anti-tumor effects were all exhibited different anti-tumor effects.The sequence inhibit rates to these 38 cases glioma cells were 26.32%,34.21%,44.74%,57.89%,15.79%.The positive expression of O6-methylguanine-DNA methyltransferase shows resistance to alkylating agents chemotherapy drugs.High grades glioma cells also manifested the more notable resistance.Conclusion Each chemotherapy drugs exhibited distinctive activity inhibition to glioma cells.Grade Ⅳ glioma cells had more chemotherapy drug resistance than other type.The positive expression of O6-methylguanine-DNA methyltransferase shows resistance to alkylating agents chemotherapy drugs.Flow cytometry has more sensitive than MTT,but the specificity is lower.
