摘要 目的探讨肺纤维化小鼠肺组织中基质金属蛋白酶-9(MMP-9)和转化生长因子-β1(TGF-β1)的表达及意义。方法将60例C57BL/6小鼠随机均分为实验组、治疗组和对照组,其中实验组和治疗组采用博莱霉素诱导建立肺纤维化小鼠模型,其余20例健康小鼠作为对照组,采用免疫组化SP法测定实验组和对照组肺组织中MMP-9和TGF-β1蛋白的表达情况。并采用格列卫对治疗组进行干预,对治疗前后肺组织中MMP-9和TGF-β1的表达情况进行比较。结果对免疫组化测得的光密度值进行比较,实验组MMP-9和TGF-β1的值明显高于对照组,差异有统计学意义(P<0.05)。将治疗组中进行干预的20只小鼠进行MMP-9和TGF-β1的光密度值测定,干预前后进行比较,差异有统计学意义(P<0.05)。结论肺纤维化小鼠肺组织中MMP-9和TGF-β1呈高表达,格列卫对博莱霉素诱导的小鼠肺纤维化程度可能有抑制作用。 Objective To expore the expression and significance of MMP-9 and TGF-[M in pulmonary fibrosis in mice lung tissue. Methods Through the establishment of pulmonary fibrosis model in mice, 60 cases of C57BL/6 mice were randomly divided into the experimental group, treatment group and control group. MMP-9 and TGF-β1 protein expression in lung tissue was determined by immunohistochemieal SP method. MMP-9 and TGF-β1 in the lung tissue before and after the intervention of the treatment group were compared. Results The experimental group and the control group by immunohistochemieal measured optical density value were compared, MMP-9 and TGF-β1 were significantly higher than the control group, the difference was statistically significant (P 〈 0.05). For 20 mice to intervene in the treatment group, MMP-9 and TGF-β1 optical density value were measured, before and after intervention were compared, the difference was statistically significant (P 〈 0.05). Conclusion The expression of MMP-9 and TGF-β1 in pulmonary fibrosis in mice is high. Gleevec for Bleomycin-induced pulmonary fibrosis may have inhibitory effect.
