猫汉赛巴尔通氏体的ELISA检测方法的建立与应用

Establishment and application of ELISA method based on outer membrane protein of cat Bartonella henselae

为提高猫汉赛巴尔通氏体（B．henselae）病血清学诊断方法的便捷性、特异性与敏感性，本研究采用纯化的原核重组表达的B．henselae外膜蛋白作为包被抗原建立了间接ELISA方法。经特异性试验、敏感性试验、重复性试验表明，该方法特异性与敏感性较高、批内与批间变异系数分别控制在0．65％～3．93％和0．63％～3．5％。经临床应用试验显示，上海近郊地区B．henselae血清阳性率为41.35％，其中流浪猫感染率差异极显著（x2=13．85、p〈0．01），老龄宠物猫的感染率差异显著（寿7．10、p〈0．05）。该方法易于操作，具有较好的应用前景，对猫B．hen．selae病的诊断、防控及公共卫生安全均具有重要意义。

To develop the diagnostic method for Bartonella henselae infection in cat, the indirect ELISA was established with the purified outer membrane protein of B.henselae. Under the optimized reaction conditions, the indirect ELISA was high specificity and sensitivity for detection of the antibody against B.henselae. The intra-assay and inter-assay coefficient of variation were 0.65 to 3.93% and 0.63 to 3.5%, respectively. In addition, the assay was applied to a survey for detection of B.henselae infection in cat on suburban of Shanghai areas, the seropositive rate was 41.35%, of which the infection in stray cats was extremely higher （x2=-13.85, p〈0.01） and infection in older pet cats was significant higher （x2=-7.10, p〈0.05）. These results demonstrated the indirect ELISA had potential to be used for application in the diagnosis of B.henselae,infections.
