Preparation of Alantolactone and Isoalantolactone from Inula racemosa Hook.f.

摘要 建立了藏木香中土木香内酯和异土木香内酯的制备方法。藏木香药材粗粉用石油醚浸泡,经硝酸银-硅胶柱色谱吸附,以苯:石油醚:乙酸乙酯(15:3:3)洗脱,得到土木香内酯和异土木香内酯的粗品,经制备型高效液相色谱分离纯化。采用DAC-HB50 C_(18)动态轴向压缩柱,以乙腈:0.1%磷酸(55:45)洗脱,检测波长194 nm。结果土木香内酯和异土木香内酯纯度达到99.1%和99.4%。 A preparation method of alantolactone and isoalantolactone from Inula racemosa Hook.f. was established. The root of Inula racernosa Hook.f. was soaked in petroleum ether, and the concentrated solution was separated on a silver nitrate-silica gel column by eluting with the mobile phase of benzene : petroleum ether : ethyl acetate (15 : 3 : 3). The crude products of alantolactone and isoalantolactone were collected and purified by preparative HPLC. The DAC-HB50 C18 column was used with the mobile phase of acetonitrile : 0.1% phosphoric acid (55 : 45) at the detection wavelength of 194 rim. The purities of alantolactone and isoalantolactone were 99.1% and 99.4 % by analytic HPLC, respectively.
