青天葵黄酮F的抗炎作用 被引量:7

Anti-inflammatory Effect of Nervilifordin F

摘要 RAW264.7细胞分别用阳性药地塞米松和青天葵黄酮F(nervilifordin F,从毛唇芋兰中提取得到)处理后,再用脂多糖(LPS)刺激2 h诱导炎症反应。采用Griess法测定一氧化氮(NO)含量,ELISA法检测前列腺素E_2(PGE_2)、白介素-6(IL-6)、肿瘤坏死因子(TNF)-α的含量,Western Blotting法检测诱导型一氧化氮合酶(iNOS)、环氧合酶(COX)-2等的表达。结果表明,青天葵黄酮F能显著降低LPS诱导的RAW264.7细胞中NO、PGE_2、IL-6和TNF-α的含量,抑制iNOS和COX-2蛋白的表达(P<0.05或P<0.01)。提示该化合物可能具有一定抗炎作用。 After treated with dexamethasone (positive control) or nervilifordin F extracted from Nerviliafordii (Hance) Schltr, RAW264.7 macrophage cells were stimulated by lipopolysaccharide (LPS) for 2 h. Nitric oxide (NO) production was analyzed by Griess reaction. The secretions of prostaglandin E2 (PGE2), interleukin-6 (IL-6) and tumor necrosis factor (TNF) -α were analyzed by ELISA. The protein expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX) -2 were analyzed by Western Blotting method. The results showed that nervilifordin F could inhibit the release of NO, PGE2, IL-6 and TNF-α, as well as the protein expressions of iNOS and COX-2 in RAW264.7 cells induced by LPS (P〈0.05 or P〈0.01). It indicated that nervilifordin F might have anti-inflammatory effect.
