摘要
目的检测生长抑素受体(SSTR)亚型SSTR2、SSTR5和p-ERK在人胃癌及正常胃组织中的mRNA和蛋白表达差异,从而探讨三者之间的相关性。方法应用免疫组化方法检测23例胃癌组织和20例正常胃组织中SSTR2、SSTR5和p-ERK的蛋白表达差异,应用real time PCR技术检测三者的mRNA表达差异。结果在蛋白水平,胃癌与胃正常组织SSTR2的表达差异无显著性(P>0.05),胃癌SSTR5的表达高于胃正常组织(P<0.05),胃癌p-ERK的表达高于胃正常组织(P<0.05);在mRNA水平,胃癌与胃正常组织SSTR2的表达差异无显著性(P>0.05),胃癌SSTR5的表达高于胃正常组织(P<0.05),胃癌ERK的表达高于胃正常组织(P<0.05);胃癌中SSTR2与ERK表达无相关性(P>0.05),SSTR5与ERK表达呈正相关(P<0.05)。结论人胃癌组织中,高表达SSTR5与p-ERK,低表达SSTR2、SSTR5与ERK在胃癌的发生发展中起重要作用,且二者之间可能通过某种机制相互关联。
Objective To detect the expression of somatostatin receptor subtypes SSTR2, SSTR5 and p-ERK at the transcription and protein levels in human gastric cancer tissues, and to investigate the correlation among them. Methods Immunohistochemistry was used to detect protein expression of SSTR2, SSTR5 and p-ERK in 23 human gastric cancer tissues and 20 human normal gastric tissues, real-time PCR was used to detect mRNA expression of them. Results At the protein level, there were not statistic differences about the expression of SSTR2 between human gastric cancer and normal gastric tissues (P〉0.05), the expression of SSTR5 and p-ERK were higher in human gastric cancer tissues than in normal gastric tissues(P〈0.05);At the transcription level, there were also not statistic differences about the expression of SSTR2 between human gastric cancer and normal gastric tissues (P〉0.05), the expression of SSTR5 and p-ERK were higher in human gastric cancer tissues than in normal gastric tissues(P〈0.05);there was no correlation between SSTR2 and ERK, there was a positive correlation between SSTR5 and ERK. Conclusion In human gastric cancer tissues, there is a higher expression of SSTR5 and p-ERK and a lower expression of SSTR2;SSTR5 and ERK play a important part in the generation and progress of human gastric cancer. There exists correlation between SSTR5 and activation of ERK through some signal pathways.
