摘要
通过对NaHCO3胁迫后刚毛柽柳(Tamarix hispida)7个转录组分析,鉴定获得了一个叶绿素a/b结合蛋白基因(命名为Thcab1)全长cDNA序列,该基因全长1 293 bp,编码区长822 bp,编码含273个氨基酸的蛋白质,编码蛋白的分子量为29.44 ku,理论等电点为7.84,NaHCO3胁迫不同时间点转录组数据表明,Thcab1基因的表达主要表现为受盐碱胁迫抑制。为了研究该基因对NaCl胁迫不同时间的应答情况,采用实时定量RT-PCR技术分析刚毛柽柳在NaCl胁迫处理后不同时间、不同组织中基因的表达。结果表明,在刚毛柽柳根和叶中,NaCl胁迫24 h内Thcab1基因主要表现为上调表达。
The full length cDNA of a light-harvesting chlorophyll a/b-binding protein ( named Thcab1 ) gene was isolated from seven transcriptome cDNA librarys of Tamarix hispida under NaHCO3 stress.The Thcab1 cDNA was 1 293 bp, and open reading frame (ORF) was 822 bp, encoding a protein of 273 amino acid residues with the molecular mass was 29.44 ku and theoretical pI was 7.84.The results of transcriptomes show that Thcab1 is obvious down-regulated under NaHCO3 stress.The expressions of Thcab1 in T.h ispida responding to NaCl in different tissues at diffident period are further inves-tigated by using quantitative RT-PCR.Thcab1 is induced in roots and leaves of T.hispida during 24 hours under NaCl stress.
