响应面法优化山稔叶总黄酮提取工艺的研究 被引量:9

Optimization of extracting total flavonoids from leaves of Rhodomyrtus tomentosa(Ait.) Hassk by response surface methodology

摘要 在提取温度、乙醇体积分数、液固比、提取时间4个单因素实验基础上,利用响应面分析法对回流法提取山稔叶总黄酮的最佳工艺进行了研究。结果表明,最优提取条件为提取温度73℃、乙醇体积分数50%、液固比19∶1(mL/g)、提取时间124min,总黄酮提取率为5.426%,与模型预测值基本相符。山稔叶总黄酮对DPPH自由基清除作用呈一定的量效关系,其半数抑制浓度为9.797μg/mL。 Based on single factor experiments, extraction conditions concluding extraction temperature, ethanol concentration, liquid-solid ratio and extraction time were optimized using response surface methodology. The results showed that the optimal extraction conditions were as follows: extraction temperature 73℃ ,ethanol concentration 50%, liquid-solid ratio 19: 1 (mL/g) and extraction time 124min, resulting in an extraction efficiency of 5.426% for total flavonoids which was consistent with the predicted value. In addition, total flavonoids from leaves of R. tomentosa had scavenging effect on DPPH with a dose -effect relationship, and the ICEO was 9.797μg/mL.
