上皮型钙黏蛋白在小鼠皮肤创伤愈合再上皮化过程中的表达 被引量:2

摘要 目的探讨上皮型钙黏蛋白在小鼠皮肤创伤愈合再上皮化过程中的表达特点和意义。方法将15只C57小鼠,在背部正中手术形成直径3mm全层性皮肤切除创口,分别在第0天、第5天和第10天处死小鼠各5只,收集取材背部创伤区皮肤组织,行HE染色,E-cadherin和K10的免疫荧光检测并进行半定量分析。结果常规HE染色结果显示小鼠第5天时创口未闭合,至第10天创口闭合。E-cadherin和K10的免疫荧光染色结果显示在第0天的小鼠表皮组织中弥漫高强度表达,至第5天在再上皮化表皮中表达减低,第10天恢复较高水平表达。E-cadherin和K10半定量检测结果显示第5天组明显低于第0天和第10天(P<0.05)。结论上皮型钙黏蛋白在小鼠皮肤创伤愈合再上皮化过程中呈动态下调表达模式,上皮型钙黏蛋白的动态下调可能是通过降低角化形成细胞间黏着连接形成并增强再生表皮的移行能力,参与对小鼠皮肤创伤愈合再上皮化过程的调节作用。 Objective To investigate the expression pattern of E- cadherin in the re- epithelialization epidermis during skin wound healing in mice. Methods The dorsal skin of 15 C57 mice was shaved,and two full- thickness excisional wounds were made in each mice on both sides in the middle of the dorsum using 3- mm biopsy punch. The mice were killed by CO 2 inhalation. Wounds were collected at indicated time points( day 0,day 5 and day 10). The wound tissues were fixed in 4% formaldehyde and then embedded in paraffin. 5- μm tissue sections from the center and stained with hematoxylin / eosin or immunofluorescent double staining of E- cadherin and K10. The semiquantitative analysis of E- cadherin and K10 were performed by Metamorph software. Results For HE staining,the wounds were not closure at day and fully closure at day 10. For immunofluorescent,E- cadherin and K10 were diffusedly and strongly expressed in the epidermis of the skin at day 0 in the mice. At day 5,the expression of E- cadherin and K10 was significantly decreased in the wound edge epidermis. At day 10,the expression of E- cadherin and K10 was recovered to a relative high intensity which was similar with the epidermis of day 0. The result of semiquantitative analysis of E- cadherin and K10 were showed that the fluorescent intensity of day 5 was significantly lower than that of the day 0 and day 10 group( P 0. 05). Conclusion E- cadherin expression is dynamically regulated during wound healing process of mice. Down regulation of E- cadherin might be involved in the regulation of migration and maturation of the re- epithelialization epidermis of mice.
